No, all we're checking here is basically the continuity of the cable which extends from the battery to the alternator, to make sure it's intact and/or not corroded. The key being on has no effect on this test. Instead of hitting the positive probe of the ohmmeter directly to the postive battery post, you touch the end of the cable at the alternator, but if the battery-to-solenoid and solenoid-to-alternator cables are good, the voltage reading should be the same.
